Epigenetic Regulation:From Code to Landscape

Abstract

In eukaryotic organisms, access to genetic information is controlled by the dynamic ON/OFF states of chromatin. Over the past decades, a remarkable diversity of covalent modifications on histones and DNA has been identified. These chemical modifications provide vast indexing potential across the genome and constitute an additional layer of epigenetic information, often referred to as the “histone or epigenetic code,” which governs the flow of genetic information at the chromosomal level. The establishment, maintenance, and interpretation of these chromatin marks are mediated by chromatin “writers,” “erasers,” and “readers” operating within highly coordinated epigenetic regulatory pathways. In this presentation, I will discuss the molecular mechanisms underlying the recognition and catalysis of hallmark epigenetic modifications by epigenetic regulators. Structural and functional studies reveal how modular protein domains selectively recognize or catalyze specific histone and DNA modifications and assemble into regulatory complexes that translate these chemical signatures into functional outcomes. Together, these mechanisms illustrate how the epigenetic code is interpreted to shape chromatin states and the broader epigenetic landscape governing gene expression programs and cellular identity.

李海涛,清华大学基础医学院院长、教授。1997年毕业于山东大学,2003年获中国科学院生物物理研究所博士学位。随后在美国纪念斯隆–凯特琳癌症中心从事博士后研究,并于2006年晋升为高级研究科学家。2010年加入清华大学,2016年获长聘教授。长期从事表观遗传学研究,先后发现并阐明以PHD、YEATS结构域为代表的一系列表观元件识别或催化组蛋白/DNA修饰的分子基础及其功能。已发表论文140余篇,被引用近2万次。曾获谈家桢生命科学创新奖、国家杰出青年科学基金、长江学者特聘教授、北京市自然科学二等奖等荣誉或奖励。
